The Source of the Amazon is a mystic place - not only because it is the origin of the biggest river system on earth but because the exact location is a fairly recent discovery by Loren McIntyre in 1971. Unlike most other rivers the sheer amount of amazon tributaries is simply so immense that the furthest source remained a mystery for so long. It is a good idea to take a guide when doing this trek part of the way is actually "off-track" in a high desert and the exact location of the source(s) is far from easy to find. There`re surely many variants how to do this trek but a typical tour lasts about 4 days/3 nights. The route starts and ends at or near the Colca Canyon - one of the deepest canyons and in fact deeper than its more famous cousin in the USA. Typical for many treks in the Andes the biggest problem is the altitude. On our tour variant we started at 4700m with two mountain passes beyond 5100m. So the gain in altitude is actually fairly moderate but do yourself a favor and acclimatize before you go for it. Please note that you also have to sleep on that level for two nights! That's not for everyone actually (we had a girl who aborted the vacation after this trek). Technically the route is very easy.
